沉默EGFR对脑出血模型大鼠学习记忆能力及海马区细胞凋亡的影响

摘要:
目的 探讨沉默EGFR对脑出血模型大鼠学习记忆能力及海马区细胞凋亡的影响。方法 选取40只SD 健康雄性大鼠,随机选取10只作为正常组,其余30只大鼠建立脑出血模型并分为沉默组、模型组、对照组。正常组、模 型组大鼠侧脑室注射8 pL PBS,对照组大鼠侧脑室注射8 pL空病毒载体,沉默组大鼠侧脑室注射8 pL EGFR siRNA 慢病毒表达载体.对各组大鼠学习记忆能力进行测试,MTT检测各组大鼠海马区细胞增殖能力,流式细胞仪检测各组 大鼠海马区细胞凋亡情况,Western blot法检测Bel 2、Bax、caspase3相对表达量。结果 沉默组大鼠逃避潜伏期长于正 常组,短于模型组、对照组,穿越平台次数低于正常组,高于模型组、对照组(P<0. 05);沉默组大鼠各时间点海马区细胞 增殖率均低于正常组,高于模型组、对照组(P<0.05);沉默组大鼠各时冋点海马区细胞凋亡率均高于正常组,低于模型 组、对照组(P<0. 05);沉默组大鼠Bel 2^caspaseS相对表达量低于正常组,高于模型组、对照组,Bax相对表达量高于正 常组,低于模型组、对照组(P<0. 05)。结论 沉默EGFR能够提升脑出血大鼠学习记忆能力,调控细胞凋亡相关蛋白、 Bel 2、Bax、caspase3相对表达量,从而促进脑出血大鼠海马区细胞增殖,抑制脑出血大鼠海马区细胞凋亡。

Abstract:
Objective To investigate the effects of silencing EGFR on learning and memory ability and apoptosis of hippocampal cells in rats with cerebral hemorrhage. Methods 40 SD healthy male rats were selected. 10 rats were randomly selected as the normal group. The other 30 rats were divided into silence group, model group and control group. In normal group and model group, 8 p 1 PBS was injected into the lateral ventricle, 8 p 1 CAV vector was injected. into the lateral ventricle of control group ? and 8 卩 1 EGFR siRNA lentivirus expression vector was injected into the lateral, ventricle of silence group. The ability of learning and memory of rats in each group was tested. The proliferation of hipp ocampal cells in each group was detected by MTT. The apoptosis of hippocampal cells in each group was detected by flow cytometry. The relative expression of bcl 2, Bax and caspase 3 was detected by Western blot. Results The escape latency of silent group was longer than that of normal group, shorter than that of model group and control group. The number of times of crossing platform was lower than that of normal group ? higher than that of model group and control group (P<0. 05). The proliferation rate of hippocampal cells in silent group was lower than that in normal group at each time point, higher than that in model group and control group (P<0. 05). The apoptotic rate of hippocampus cells in silent group was higher than that in normal group, lower than that in model group and control group (P<0.05), The relative exp res sion of Bcl 2 and Caspase 3 in silent group was lower than that in normal group, higher than that in model group and con trol group, and the relative expression of Bax in silent group was higher than that in normal group, lower than that in model group and control group (P<0.05). Conclusion Silencing EGFR can improve the learning and memory ability of rats with cerebral hemorrhage, regulate the relative expression of apoptosis related proteins? B.cl 2, Bax and caspase 3., so as to promote the proliferation of hippocampal cells in rats with cerebral hemorrhage and inhibit the apoptosis of hipp ocampal cells in rats with cerebral hemorrhage.
