反复呼吸道感染患儿外周血lncRNA NEAT1的表达及与Th1/Th2平衡的关系

【摘要】 目的 探讨长链非编码RNA核富集转录体1(LncRNA NEAT1)在反复呼吸道感染(RRI)患儿血浆中的表达情况,并分析其与Th1/Th2平衡的关系。 方法 选取2018年6月~2019年3月本院收治的68例RRI患儿为RRI组,并选取同期74例健康幼儿为健康组。采用实时荧光定量PCR(qRT-PCR)法检测血浆lncRNA NEAT1水平;以流式细胞仪检测Th1、Th2细胞占CD4+T细胞的比例,计算Th1/Th2比值;采用酶联免疫吸附法(ELISA)检测血浆干扰素-γ(IFN-γ)、白介素-4(IL-4)水平;观察RRI患儿血浆lncRNA NEAT1、Th1、Th2、Th1/Th2水平与IFN-γ、IL-4,及lncRNA NEAT1表达水平与Th1/Th2平衡的关系。 结果 与健康组相比,RRI组患儿血浆lncRNA NEAT1表达水平、外周血CD4+Th2细胞比例、IL4水平明显升高(t=9.198、10.218、27.449,P均<005),外周血CD4+Th1细胞比例、Th1/Th2比值、IFN-γ水平明显降低(t=10.689、14.572、23.665,P均<0.05);RRI组患儿血浆lncRNA NEAT1表达水平与IFNγ呈负相关(r=〖CD*3/5〗0383,P<0.05),Th1/Th2比值与IL-4、lncRNA NEAT1表达水平与Th1/Th2比值均呈负相关(r=〖CD*3/5〗0.428,P<0.05),Th1细胞比例、Th1/Th2比值与IFN-γ呈正相关(r=0.458、0.327,P均<0.05),lncRNA NEAT1表达水平、Th2细胞比例与IL-4(r=0.484、0.537,P均<0.05)呈正相关。 结论 RRI患儿血浆lncRNA NEAT1呈高水平,Th1/Th2比值呈低水平,lncRNA NEAT1与Th1/Th2有一定负相关性,两者可能相互作用,从而共同影响RRI发生发展。

【Abstract】 Objective To observe the expression of long noncoding RNA nuclear enriched abundant transcript 1 (lncRNA NEAT1) in the plasma of children with recurrent respiratory tract infection (RRI) and analyze its relationship with Th1/Th2 balance. Methods From June 2018 to March 2019, 68 children with RRI in our hospital were selected as RRI group, and 74 healthy children were selected as health group. The level of plasma lncRNA NEAT1 was detected by realtime fluorescence quantitative PCR (qRTPCR). The percentages of Th1 and Th2 cells to CD4 + T cells were measured by flow cytometry. The ratio of Th1/Th2 was calculated. The levels of interferonγ (IFN-γ) and interleukin4 (IL-4) were detected by enzymelinked immunosorbent assay (ELISA). The relationships between the levels of lncRNA NEAT1, Th1, Th2, Th1/Th2 and IFN-γ, IL-4 and between the expression level of lncRNA NEAT1 and Th1/Th2 balance were observed. Results The expression level of plasma lncRNA NEAT1, the percentage of CD4 + Th2 cells and the level of IL-4 in RRI group were significantly higher than those in healthy group (t=9.198, 10.218, 27.449, all P<0.05). The percentage of CD4 + Th1 cells, ratio of Th1/Th2 and level of IFN-γ in peripheral blood decreased significantly (t=10.689, 14.572, 23665, all P<0.05). There were negative correlations between the expression level of plasma lncRNA NEAT1 and IFNγ (r=〖CD*3/5〗0383, P<0.05), between Th1/Th2 ratio and IL-4 (r=〖CD*3/5〗0.428, P<0.05), between the expression level of lncRNA NEAT1 and Th1/Th2 ratio in children with RRI (r=〖CD*3/5〗0.428, P<0.05), and there were positive correlations between percentage of Th1 cell, Th1/Th2 ratio and IFNγ (r=0.458, 0.327, both P<0.05), between the expression level of lncRNA NEAT1, percentage of Th2 cell and IL-4 (r=0.484, 0.537, both P<0.05). Conclusion The plasma lncRNA NEAT1 level of RRI children is high. The ratio of Th1/Th2 is low. There is a negative correlation between lncRNA NEAT1 and Th1/Th2, which may interact with each other, thus jointly affect the occurrence and development of RRI.
