LncRNA PVT1 miR-30d表达与上皮性卵巢癌患者化疗敏感性及预后的关系

【摘要】 目的 探讨LncRNA PVT1、miR30d表达与上皮性卵巢癌患者化疗敏感性及预后的关系。 方法 选取2012年2月~2014年1月行手术治疗的76例上皮性卵巢癌患者的癌组织标本作为上皮性卵巢癌组,另选取本院收治的76例良性卵巢肿瘤作为对照组。检测LncRNA PVT1、miR30d相对表达量,根据表达水平分为高表达组与低表达组;根据对顺铂(DDP)、阿霉素(ADM)、环磷酰胺(CTX)的敏感性将患者分为敏感组和耐药组并对比LncRNA PVT1、miR30d表达情况;应用Pearson法对miR30d和LncRNA PVT1表达的相关性进行分析;采用KaplanMeier对上皮性卵巢癌患者5年的生存情况进行分析。 结果 与对照组相比,上皮性卵巢癌组miR30d表达水平降低(P<0.05),LncRNA PVT1表达水平升高(P<0.05)。miR30d、LncRNA PVT1表达均与FIGO分期、分化程度密切相关(P<0.05)。DDP、ADM敏感组miR30d表达水平显著高于耐药组(P<005),LncRNA PVT1表达水平显著低于耐药组(P<0.05)。Pearson相关性分析显示上皮性卵巢癌组织中miR30d和LncRNA PVT1表达呈负相关(P<005)。KaplanMeier法分析显示miR30d高表达组5年内总生存率及中位生存时间远高于低表达组(73.7% VS 33.3%,28.0月 VS 20.0月,P<0.05);LncRNA PVT1低表达组5年内总生存率及中位生存时间远高于高表达组(81.5% VS 22.4%,27.0月 VS 18.0月,P<0.05)。 结论 上皮性卵巢癌患者癌组织miR-30d低表达,LncRNA PVT1高表达,miR-30d、LncRNA PVT1表达与上皮性卵巢癌患者化疗敏感性及预后有关。

Relationships between expressions of LncRNA PVT1 and MiR-30d with chemosensitivity and prognosis of epithelial ovarian cancer patients

【Abstract】 Objective To study the abnormal expressions of long noncoding RNA PVT1 (LncRNA PVT1) and micro RNA30d (miR30d) in epithelial ovarian cancer, and explore the relationships between LncRNA PVT1 and miR30d with chemosensitivity and prognosis of patients with epithelial ovarian cancer. Methods 76 patients with epithelial ovarian cancer underwent surgical treatment from February 2012 to January 2014 were selected as the epithelial ovarian cancer group, and 76 patients with benign ovarian tumors in our hospital were selected as the control group. The relative expressions of LncRNA PVT1 and miR30d were detected by qRTPCR. According to the sensitivity to Cisplatin (DDP), Adriamycin (ADM), Cyclophosphamide (CTX), the patients were divided into sensitive group and resistant group, and the expressions of LncRNA PVT1 and miR30d were compared. Pearson method was used to analyze the correlation between the expressions of MV30d and LncRNA PVT1. KaplanMeier was used to analyze the 5year survival of patients with epithelial ovarian cancer. Results Compared with the adjacent tissue group, the expression of miR30d in epithelial ovarian cancer group decreased (P<0.05), while the expression of LncRNAPVT1 increased (P<0.05). The expressions of miR30d and LncRNAPVT1 were closely related to FIGO stage and differentiation degree. The expression level of miR30d in DDP and ADM sensitive group was significantly higher than that in drug resistant group, and the expression level of LncRNAPVT1 was significantly lower than that in drug resistant group (P<0.05). Pearson correlation analysis showed that the expressions of MV30d and LncRNA PVT1 were negatively correlated in epithelial ovarian cancer (P<0.05). KaplanMeier analysis showed that the 5year overall survival rate and median survival time of the miR30d highexpressed group were significantly higher than those of the lowexpressed group (73.7% VS 33.3%, 2810months VS 2010months, P<0.05). The 5year overall survival rate and median survival time of LncRNA PVT1 lowexpressed group were much higher than those of highexpressed group (81.5% VS 22.4%, 2710months VS 1810months, P<0.05). Conclusion In cancer tissues of patients with epithelial ovarian cancer, the expression of miR30d is low and LncRNA PVT1 is high. The expressions of miR30d and LncRNAPVT1 are related to the chemosensitivity and prognosis of epithelial ovarian cancer.
