沉默环状RNA hsa_circ_0011021表达对宫颈癌细胞增殖 迁移及侵袭的影响

摘要:
【摘要】 目的 探讨环状RNA(circRNA)hsa_circ_0011021在宫颈癌组织和细胞株中的表达变化及其对宫颈癌细胞增殖、迁移和侵袭能力的影响。方法 选取河南科技大学第一附属医院就诊的15例宫颈癌患者,取15对宫颈癌组织和癌旁组织。qRTPCR实验检测hsa_circ_0011021在宫颈癌组织和宫颈癌癌旁组织(Hela和SiHa)细胞中的相对表达水平;CCK8和克隆形成实验检测沉默hsa_circ_0011021对Hela细胞活性和克隆形成能力的影响;划痕愈合实验和Transwell侵袭实验检测沉默hsa_circ_0011021对Hela细胞迁移及侵袭能力的影响。结果 hsa_circ_0011021在宫颈癌组织的相对表达水平明显高于癌旁组织(P<0.05);hsa_circ_0011021在Hela和SiHa细胞中的相对表达水平明显高于人永生化表皮细胞HaCaT(P<0.05)。在Hela细胞沉默hsa_circ_0011021,Hela细胞的活性和克隆形成能力显著降低,迁移距离明显减少,侵袭能力也明显下降(P<0.05)。结论 hsa_circ_0011021在宫颈癌组织和宫颈癌细胞(Hela和SiHa)中的表达水平明显升高;沉默hsa_circ_0011021显著抑制宫颈癌Hela细胞的增殖、迁移和侵袭。

Abstract:
【Abstract】 Objective To explore the expression of the circular RNA (circRNA) hsa_circ_0011021 in cervical cancer tissues and cell lines, and study the effect of hsa_circ_0011021 knockdown on the proliferation, migration and invasion of cervical cancer cells. Methods The expression levels of hsa_circ_0011021 in cervical cancer tissues and cervical cancer cell lines (Hela and SiHa) were determined by quantitative realtime PCR (qRTPCR). The effect of hsa_circ_0011021 on the viability and proliferation of cervical cancer cells, CCK8 and colony formation assays were performed in Hela cells were observed. Transwell migration assay and wound scratch assay were carried out to evaluate the effect of hsa_circ_0011021 on the migration and invasion ability of Hela cells.Results The expression level of hsa_circ_0011021 in cervical cancer tissues and Hela and SiHa cells was higher than that in adjacent normal tissues and human immortal keratinocyte line (HaCaT). Knockdown of hsa_circ_0011021 obviously inhibited the proliferation, migration and invasion of Hela cells. Conclusion Knockdown of hsa_circ_0011021 remarkably suppresses the proliferation, migration and invasion of cervical cancer cells.
