DNA甲基化在结直肠癌中的研究进展

摘要:
【摘要】 结直肠癌(colorectal cancer,CRC)是使结肠上皮细胞转变为侵袭性的恶性腺瘤,其过程主要由多基因组学累积和表观遗传学改变导致的。表观遗传学是指研究基因的核苷酸序列不发生改变的情况下,使基因表达的程度发生变化,囊括DNA甲基化、组蛋白修饰、染色质重塑及RNA干扰等。其中DNA甲基化是最重要的一种表观遗传修饰,亦然是当前研究最深入的领域。有研究发现,表观遗传学的改变在结直肠癌中发挥着重要的作用,在癌变的过程中,以上一系列表观遗传学的改变通常会使癌基因获得功能或抑癌基因丢失功能。本文就最近几年结直肠癌DNA甲基化相关性研究的进展进行综述。

Research development of DNA methylation in colorectal cancer

Abstract:
【Abstract】 Colorectal cancer (CRC) is caused by multiple genomic accumulation and epigenetic changes that transform colonic epithelial cells into aggressive malignant adenomas. Epigenetics refers to the study of changes in gene expression levels, including DNA methylation, histone modification, chromatin remodeling, and RNA interference, when nucleotide sequences of genes are not altered. Among them, DNA methylation is the most important epigenetic modification, and it is also the most indepth field of study. Studies have found that epigenetic changes play an important role in the carcinogenesis of colorectal cancer, in these processes, some of the above list of epigenetic change typically have the function or loss of tumor suppressor gene function to cancer gene. This article reviews the recent advances in the study of DNA methylation in colorectal cancer.
