UVB诱导人皮肤成纤维细胞改变后β-catenin基因表达水平的研究

摘要:
【摘要】 目的 研究经过不同剂量紫外线中波(UVB)照射正常人皮肤成纤维细胞(HSF) 不同时间后β链蛋白(βcatenin)基因的表达改变,探讨UVB诱导HSF衰老可能的机制。方法 未经UVB照射的HSF为对照组;经过不同剂量(100、200、300) mJ/cm2 UVB处理不同时间(1、2、3)天后的HSF为实验组。实时荧光定量PCR技术(FQPCR)检测实验组和对照组中β-catenin基因的表达;四唑盐比色法(MTT)观察HSF的生长活性改变,流式细胞仪检测HSF的凋亡率变化。结果 UVB照射对HSF细胞的生长有明显抑制作用,并且随着UVB照射时间延长和剂量的增加,HSF细胞生长率逐渐下降;UVB处理后实验组中β-catenin基因的相对表达水平明显低于对照组,差异具有统计学意义(P<005);β-catenin基因的表达水平随着UVB作用的时间和剂量的不同而变化,存在较为明显的时间依赖性和剂量依赖性。结论 经过UVB处理的HSF细胞中调控皮肤衰老的基因β-catenin表达明显降低,大剂量长时间UVB照射可诱导HSF发生衰老改变。

Abstract:
【Abstract】 Objective To study the expression of βcatenin in HSF after different dose and time treatments with UVB, and explore the mechanism of UVB to induce HSF senescence. Methods The control group was not treated with UVB. The experimental group was treated with UVB (100, 200 , 300 μmJ/cm2) for different time (1, 2, 3d). The expression of βcatenin gene was detected with FQPCR. The growth activity of HSF was observed by MTT. The apoptosis rate of HSF was detected by flow cytometry. Results HSF treated with UVB displayed a slow growth in comparison with the control group, and the growth rate decreased with the creasing of UVB dose. The expression level of βcatenin gene in the experimental group was significantly lower than that in the control group, the difference was statistically significant(P<005). The expression level of the genes varies with the time and dose of UVB, and it has obvious timedependent and concentrationdependent. Conclusion The expression of βcatenin in the HSF treated by UVB was significantly decreased, and UVB could induce the senescence of HSF.
