不同病理类型及分期甲状腺癌中miR221 miR222 miR197和miR346的表达研究

【摘要】目的 探讨不同病理类型及分期甲状腺癌中miR221、miR222、miR197和miR346的表达水平,为临床手术方式选择及术后治疗提供参考。方法 选取2013年6月~2017年8月行甲状腺手术的40例甲状腺癌(乳头状癌、滤泡癌各20例)及癌旁正常甲状腺组织、20例良性甲状腺疾病组织,术中取材后,立即放入液氮中保存后转入-70℃保存。采用定量RTPCR检测miR221、miR222、miR197和miR346在相应组织标本中的表达水平。结果 对甲状腺癌,miR221与miR222的表达与患者的年龄、性别、最大肿瘤灶等无明显相关性(P>005),与TNM分期及淋巴转移情况相关(P<005);miR197和miR346的表达在20例甲状腺滤泡癌中均有明显升高(P<005),乳头状癌组织中未见显著变化(P>005)。miR221与miR222在良性甲状腺疾病组织表达量变化均不明显(P>005);但在甲状腺癌组织中的表达变化显著,差异均有统计学意义(P<005);对miR197和miR346在相应组织标本中的表达水平进行比较结果显示,良性甲状腺疾病组织中miR197和miR346表达没有明显变化(P>005);在20例滤泡癌组织中明显升高,差异有统计学意义(P<005);在乳头状癌组织及癌旁正常组织中的均无明显变化(P>005)。结论 不同病理类型及分期甲状腺癌中miRNA的表达可作为甲状腺癌早期诊断、淋巴结转移、复发及远处转移密切相关的预警分子,为甲状腺癌手术方式选择及术后指导治疗提供了重要依据。

Study on the expression of miR221, miR222, miR197 and miR346 in different pathological types and stages of thyroid carcinoma

【Abstract】 Objective To analyze and discuss the expression of miRNA in different pathological types and stages of thyroid carcinoma, and provide reference for the clinical operation and postoperative treatment. Methods 40 patients of thyroid carcinoma (including 20 cases of papillary carcinoma) were collected from people's Hospital of Inner Mongolia Autonomous region and affiliated Hospital of Inner Mongolia Medical University. 20 cases of follicular carcinoma) and 20 cases of benign thyroid disease tissues were collected during operation. After being stored in liquid nitrogen immediately, it was transferred to70 ℃ for storage. The quantitative RTPCR was used to detect miR221 and miR222. The expression level of miR197 and miR346 in corresponding tissues.Results For thyroid cancer, miR221 and miR222 expression and patient age and gender, had no significant correlation between the maximum tumor foci. They were correlated with TNM staging (P<005) and lymph node metastasis (P<005). miR197 and miR346 expression significantly increased in thyroid follicular carcinoma (P<005). The expression of miR221 and miR222 in benign thyroid diseases did not change significantly (P>005). However, there was a significant change in the expression of PTNE in thyroid carcinoma, There was a negative correlation between them and the expression of PTNE (P< 005). The difference was statistically significant (P< 005). The expression levels of miR197 and miR346 in corresponding tissues were compared. There was no significant change in the expression of miR197 and miR346 in benign thyroid diseases. In thyroid carcinomas 2 There was a significant increase in 0 cases of follicular carcinoma, the difference was statistically significant (P< 005), and there was no significant change in the tissues of papillary carcinoma and normal tissues adjacent to the carcinoma.Conclusion The expression of miRNA in different pathological types and stages of thyroid carcinoma is closely related to early diagnosis, lymph node metastasis, recurrence and distant metastasis of thyroid carcinoma.
