GLP-1重组减毒沙门菌治疗对2型糖尿病小鼠的

摘要:
【摘要】 目的 探讨GLP1重组减毒沙门菌治疗对2型糖尿病小鼠的Caveolin1、胰岛β细胞自噬相关因子的影响。方法 将纳入的40只小鼠分为正常组、对照组、空载组及治疗组,每组各10只。正常组正常饲养,对照组、空载组及治疗组使用链脲佐菌素(STZ)造模; 对照组及正常组采用生理盐水干预,空载组采用空载体重组减毒沙门菌干预,治疗组采用GLP1重组减毒沙门菌干预;在造模前、造模成功后及治疗结束后进行眼眶取血,使用血糖仪检测血中血糖水平,治疗结束后检测小鼠Caveolin1、胰岛β细胞自噬相关因子表达情况。结果 治疗组小鼠血糖水平治疗后显著低于对照组及空载组(P<005);治疗组小鼠胰岛Caveolin1水平治疗后显著高于对照组及空载组(P<005),对照组及空载组Caveolin1水平与治疗前比较差异无统计学意义(P>005);治疗后治疗组小鼠胰岛Atg3、Atg12及Beclin1水平显著低于对照组及空载组(P<005),对照组及空载组Atg3、Atg12及Beclin1水平较治疗前无明显差异(P>005)。结论 采用GLP1重组减毒沙门菌治疗后可有效改善2型糖尿病小鼠Caveolin1及胰岛β细胞自噬相关因子表达,有较广的应用价值。

【Abstracts】 Objective Investigate the effect of recombinant salmonella typhimurium (GLP1) on caveolin1 and βcell autophagy related factors in type 2 diabetic mice. Methods 40 mice were divided into normal group, control group, empty vector group and treatment group. The c control group, empty vector group and treatment group were treated with streptozotocin for model. The normal group and control group were treated with saline. The empty vector group was treated with attenuated Salmonella. The treatment group was treated with GLP1 recombinant attenuated Salmonella. The blood samples were taken at the end before modeling, after modeling and after treatment. The blood glucose level were detected with blood glucose meter. Results After treatment, blood glucose level was significantly lower than that of the control group and empty vector group (P<005). The islet Caveolin1 level of the treatment was significantly higher than that of the control group and empty vector group (P<005). There was no significant difference between the Caveolin1 group and control vector group (P>005). Atg3, Atg12 and Beclin1 of mouse islets of treatment were significantly lower than that of control group and empty vector group (P<005). There were no significant difference between the group and the group of Atg3, Atg12 and Beclin1 load level control (P>005). Conclusion GLP1 recombinant attenuated Salmonella can effectively improve the expression of Caveolin1 and βcell autophagy related factors in type 2 diabetic mice, and has a wide range of practical value.
