氟对SD大鼠分泌期牙胚中UCH L1表达的影响

摘要:
【摘要】 目的 观察不同浓度氟化物对分泌期大鼠牙胚中泛素羧基末端水解酶L1(UCHL1)表达的影响,为进一步研究氟牙症发生的机制提供实验依据。方法 将9只妊娠10 天的大鼠,分为3组,分别饲以去离子水和含氟浓度为50mg/L、200 mg/L的氟化钠水溶液,妊娠21天时,取其子鼠下颌骨制作切片及提取牙胚组织,分别用免疫组化和实时定量PCR方法检测不同染氟组大鼠牙胚细胞中UCHL1的蛋白及mRNA表达,应用ImagePro Plus 60软件对免疫组化结果进行图像分析。结果 加氟组胎鼠牙胚中的UCHL1的免疫组化染色程度较对照组减弱,其mRNA的表达量逐渐减少,高氟浓度下尤为明显。结论 氟化物作用下,分泌期牙胚中UCHL1的表达降低,可能与氟牙症的发生机制相关。

Abstract:
【Abstract】 Objective To investigate the effect of fluoride on the expression of UCHL1 during SD rat tooth germ development. Methods 9 pregnant SD rats were randomly divided into 3 groups. Rats in each group were fed with water containing fluoride of 0mg/L, 50mg/L and 200mg/L, respectively. The first mandibular molar germs of fetal rats were obtained at E21 days to make sections for detecting UCHL1 by immunohistochemically method,and to extraction of tooth germ tissue for detecting mRNA by realtime PCR. ImagePro Plus 60 software were used to analyze the results of immunohistochemistry. Results The stain of UCHL1 of the two experimental groups was at lower level than control group, especially in the high fluoride concentration group (200 mg/L). The expression of UCHL1 mRNA is also the same. Conclusion Under the action of fluoride, the expression of UCHL1 in ameloblasts of secretory phase decreased, which may be related to the mechanism of dental fluorosis.
